Abstract:
基于形式电荷(formal charge)参数设置, 用有限差分泊松-玻尔兹曼方法(FDPB)来预测蛋白质结构的稳定性. 以人的2Zn天然胰岛素及其速效突变体B9D(B9Ser→Asp), B9E(B9Ser→Glu), B9EB10D(B9Ser→Glu, B10His→Asp)和B10D(B10His→Asp)为研究对象, 研究了溶液中不同pH 值及盐离子浓度对天然胰岛素和突变体稳定性的影响, 阐明了这些突变体具有速效特性的结构和理论基础. 计算结果与实验相吻合.
