A solid phase extraction-high performance liquid chromatographic (SPE-HPLC) method was developed for the determination of phthalic acid esters and their metabolites in human urine. The conditions of pretreatment such as eluent, elution volume and elution velocity were optimized. The optimized conditions were 2 mL of acetonitrile, 2 mL of ethyl acetate and 1 mL of ethyl ester-n-hexane (1/19, V/V) at the flow rate of 1 mL/min. Under the optimized conditions, five phthalic acid esters and their monoesters were determined by reversed phase liquid chromatography. Good linear relationships were obtained in the range of 20 mu g/L to 10000 mu g/L with the correlation coefficients above 0. 99. The limits of detection were between 3. 8 mu g/L and 7. 0 mu g/L. The recoveries were in the range of 72. 2% to 102. 3% and relative standard deviations were lower than 8%. The method was applied to analyze five human urines collected from five fertile women. Except for dimethyl phthalate and diethyl phthalate, the others were found in the urines.
