基于拉曼光谱方法研究不同剪切力下的红细胞亚损伤 CSCD

摘要 :

利用共聚焦拉曼光谱技术,对人工心脏泵不同剪切应力下受到亚损伤的红细胞进行实验研究,验证拉曼光谱对红细胞亚损伤程度的评估能力,为血液损伤评价提供了一种新的思路。实验采集血红蛋白和红细胞的拉曼光谱标准谱图并进行对比分析,以确定红细胞谱图特征峰的归属。用血液剪切力试验平台对测试血样施加暴露时间为1 s,大小分别为0, 50, 100, 150, 200, 250和300 pa的剪切力。利用共聚焦拉曼仪器,在10倍长焦物镜,532 nm激光光源波长,积分时间10 s,积分次数2次,2.5 mW功率下采集剪切应力作用后的红细胞拉曼谱图。通过归一化的方法对比红细胞的拉曼谱图变化,评估红细胞亚损伤的程度,运...

Methylation signature genes identification of cancers occurrence and pattern recognition SCIE

In order to identify the signature genes of tumorigenesis, the pattern-recognition method was used to analyze the gene methylation (ME) data which included only normal and cancer samples and was collected from the TCGA (The Cancer Genome Atlas) database. Here, we analyzed the DNA methylation profiles of the six types of cancer and the ME signature genes for each cancer were selected by means of a combination of correlation, students t-test and Elastic Net. Modeling by support vector machine, the accuracy of ME signature genes can be as high as 98 % for training set and as high as 97 % for the independent test set, the recognition accuracy of stage I is more than 97 % for training set and more than 98 % for test set. Then, the common signature genes and common pathways emerging in multiple cancers were obtained. A functional analysis of these signature genes indicates that the identified signatures have direct relationship with tumorigenesis and is very important for understanding the pathogenesis of cancer and the early therapy.

Hemodynamic effects of pulsatile unloading of left ventricular assist devices (LVAD) on intraventricular flow and ventricular stress SCIE

The role of pulsatile unloading in hemodynamic changes in intraventricular flow and ventricular wall stress remains unknown. In this study, a finite element model of the left ventricle (LV) is proposed to calculate the mechanical response. The constitutive model of the LV is composed of a quasi-incompressible transversely isotropic model and an active contraction of the myocardium model. Pulsatile unloading is provided by the left ventricular assist device (LVAD), which is implanted between the aortic root and aortic arch. Support models (constant speed and co-pulse) were utilized to study the effect of pulsatile unloading on intraventricular flow and ventricular stress. The result indicates that the formation time of the vortex increases under pulsatile unloading. The area rate of high time-averaged wall shear stress (TAWSS) increased after pulsatile unloading. The area of the high oscillatory shear index (OSI) region (OSI > 0.375) was calculated for heart failure, constant speed, and co-pulse (9.9 cm(2), 9.6 cm(2), and 9.2 cm(2), respectively). The maximum value of the stress that reflects the level of stretch declined after pulsatile unloading (66.4 kPa, 30.9 kPa, and 21.3 kPa, respectively). Besides, pulsatile unloading impacts the maximum value of thickness at the ventricular wall (-0.75 mm, -1 mm, and -1.25 mm, respectively). The change ratios of the thickness are 10%, 14%, and 17%, respectively. In conclusion, pulsatile unloading contributes to the distribution of intraventricular flow and the formation time of the vortex. Co-pulse support significantly reduces the maximum value of the ventricular wall stress and the area of high stress on the ventricular wall. (C) 2019 Elsevier Ltd. All rights reserved.

Excitation parameter preferences are key factors affecting the performance of magnetic frequency mixing detection. A uniform experimental design method was used to analyze this influence. Using fuzzy theory, a comprehensive model is established for evaluating the effect of magnetic frequency mixing. A polynomial is selected as the regression function to express explicitly the correlation between the excitation parameters and the frequency-mixing effect. The excitation parameters were then optimized using genetic algorithm. Magnetic frequency mixing experiments were conducted to measure the surface hardness of some ferromagnetic materials. Frequency mixing is further enhanced under the optimal settings, resulting in an improvement in the measurement sensitivity. The results of this study support the application of the magnetic frequency mixing technique in non-destructive testing.

The magnetic frequency mixing method was investigated for fatigue damage evaluation of medium carbon steel. Two types of magnetic signal, the magnetic flux density and the tangential magnetic field, are detected and analyzed. Two parameters, the magnetic frequency mixing factor and the power law coefficient, are proposed for magnetic frequency mixing effect characterization. On-line and off-line magnetic frequency mixing experiments were performed on specimens with various degrees of fatigue. Experimental results show that the two parameters changed along with the fatigue cycles with monotonic linear regularity and both parameters can be used for fatigue damage evaluation of medium carbon steel.
