Study on Erythrocyte Sublethal Damage Under Different Shear Stress Based on Raman Spectroscopy SCIE

In this study, confocal Raman spectroscopy was used to study the Sublethal damage erythrocytes of artificial heart pump under different shear stress to verify the ability of Raman spectroscopy to evaluate the degree of erythrocyte Sublethal damage. It provides a new idea for the evaluation of blood injury. The standard Raman spectra of hemoglobin and red blood cells were collected and compared in order to determine the peak intensity of erythrocytes. The sheer force of 0, 50, 100, 150, 200, 250 and 300 pa was applied to the tested blood samples for 1 s on the blood shear test platform. Using confocal Raman instrument, the Raman spectra of erythrocytes under shear stress were collected under 10 times long focal lens, 532 nm laser light source wavelength, integration time 10 s, twice, and 2. 5 mW. Through the normalization method to compare the changes of the Raman spectrum of red blood cells, evaluate the degree of red blood cell Sublethal damage, and use the curve fitting method to fit the characteristic peak and shear stress to verify the ability of Raman spectrum to evaluate the sub-injury of red blood cells. Comparing the standard Raman spectra of hemoglobin and red blood cells, it is found that the erythrocyte spectra can reflect the internal structure of hemoglobin. The results show that Raman spectroscopy can be used to distinguish the Sublethal damage erythrocytes under different shear stress, and it is inferred that the shear stress can pass through the cell membrane and affect the internal hemoglobin structure. With the increase of shear force, the left spectral line of 1 376 cm(-1) increased. Obviously, the peak intensity of 1 549 and 1 604 cm(-1) increased, and the vibration band of oxygen concentration marker band 10 at 1 639 cm(-1) decreased. Among them, the peak strength of 1 549 cm(-1) position is the high spin band of ferrous ion. Under the action of different shear forces, the difference of peak strength is the most obvious, which has an obvious positive linear relationship with the shear stress, and the fitting effect is good. Raman spectroscopy has the advantages of simple treatment, short time, simple operation and good reproducibility can accurately detect the subtle changes in the internal structure of cells, and evaluate the degree of sub-injury of red blood cells. it makes up for the deficiency of the traditional evaluation of hemolysis, provides a new technical means for the evaluation of blood injury caused by artificial heart pump, and broadens the application field of the Raman detection method.

Hemodynamic evaluation and in vitro hemolysis evaluation of a novel centrifugal pump for extracorporeal membrane oxygenation. PubMed

The STM CP-24 I centrifugal pump is a newly developed centrifugal pump for extracorporeal membrane oxygenation equipment. This study aimed to combine hydraulic experiments, hemodynamic numerical simulations, and standard in vitro hemolysis experiments to investigate the comprehensive performance of this centrifugal pump.In vitro experiments were first done to obtain the pressure-flow data of the centrifugal pump in its working range to evaluate its hydraulic performance. Next, the commonly used clinical working points were selected as boundary conditions, and a computational fluid dynamics method was applied to evaluate its hemodynamic performance. The blood pressure distribution, blood flow fields, and high-wall-shear-stress zones in the centrifugal pump were determined as indicators for hemodynamic evaluation. Finally, standard in vitro hemolysis experiments were performed to test the blood compatibility of this centrifugal pump (n=3 blood samples). In addition, its blood compatibility was evaluated in the form of the normalized index of hemolysis (NIH).The pressure-flow curve of the centrifugal pump showed that the head pressure and flow of the centrifugal pump showed a mostly linear relationship within the whole working range. When the rotation speed of the centrifugal pump was 5,500 rpm, it achieved a hydraulic performance of 550 mmHg head pressure and 8 L/min output flow, which could meet the clinical needs of extracorporeal membrane oxygenation. Analysis of computational fluid dynamics data indicated that the centrifugal pump had excellent hemodynamic performance: even distribution of blood pressure in the pump, no blood flow stagnation zone or dead zone in the overall flow field, and secondary flows in the gap between the rotor and the volute that significantly reduced the volume of the low-blood-flow zone close to the impeller. There was no obvious high-shear-stress zone on the surface of the volute or the impeller, which will effectively reduce the risk of thrombosis. In vitro hemolysis experiments indicated that the centrifugal pump had excellent blood biocompatibility, with a NIH =0.0125±0.0022 g/100 L.The STM CP-24 I centrifugal pump has excellent hydraulic performance, a reasonable design of the hemodynamic structure of the blood pump, and excellent blood compatibility. Therefore, it can meet clinical needs.

Heart failure is one kind of cardiovascular disease with high risk and high incidence. As an effective treatment of heart failure, artificial heart is gradually used in clinical treatment. Blood compatibility is an important parameter or index of artificial heart, and how to evaluate it through hemodynamic design and in vitro hemolysis test is a research hotspot in the industry. This paper first reviews the research progress in hemodynamic optimization and in vitro hemolysis evaluation of artificial heart, and then introduces the research achievements and progress of the team in related fields. The hemodynamic performance of the blood pump optimized in this paper can meet the needs of use. The normalized index of hemolysis obtained by in standard vitro hemolysis test is less than 0.1 g/100 L, which has good hemolysis performance in vitro. Excitation parameter preferences are key factors affecting the performance of magnetic frequency mixing detection. A uniform experimental design method was used to analyze this influence. Using fuzzy theory, a comprehensive model is established for evaluating the effect of magnetic frequency mixing. A polynomial is selected as the regression function to express explicitly the correlation between the excitation parameters and the frequency-mixing effect. The excitation parameters were then optimized using genetic algorithm. Magnetic frequency mixing experiments were conducted to measure the surface hardness of some ferromagnetic materials. Frequency mixing is further enhanced under the optimal settings, resulting in an improvement in the measurement sensitivity. The results of this study support the application of the magnetic frequency mixing technique in non-destructive testing.

Biomechanical effects of the novel series LVAD on the aortic valve EI

Background and Objective: The series type of LVAD (i.e., BJUT-II VAD) is a novel left ventricular assist device, whose effects on the aortic valve remain unclear. Methods: The biomechanical effects of BJUT-II VAD on the aortic valve were investigated by using a fluidstructure interaction method. The geometric model of BJUT-II VAD was virtually implanted into the ascending aorta to generate the realistic flow pattern for the aortic valve (i.e., support). In addition, the biomechanical states of the aortic valve without BJUT-II VAD support was computed as control (i.e., control case). Results: Results demonstrated that the biomechanical effects of BJUT-II VAD were quite different from that resulting from traditional 'bypass LVAD.' Compared with those in the control case, BJUT-II VAD support could significantly reduce the stress load of the leaflet (maximum stress, 0.5 MPa in the control case vs. 0.12 MPa in the support case). Similarly, the rapid valve opening time (100 ms in the control case vs. 175 ms in the support case) and rapid valve closing time (50 ms in the control case vs. 150 ms in the support case) in the support case were obviously longer than those in the control case. Moreover, BJUT-II VAD support reduced retrograde blood flow during the diastolic phase and significantly changed the distribution of WSS of the leaflets. Conclusions: In summary, while unloading the left ventricle, BJUT-II VAD could provide beneficial biomechanical states for the aortic leaflets, thereby reducing the risk of aortic valve disease. © 2020

The role of pulsatile unloading in hemodynamic changes in intraventricular flow and ventricular wall stress remains unknown. In this study, a finite element model of the left ventricle (LV) is proposed to calculate the mechanical response. The constitutive model of the LV is composed of a quasi-incompressible transversely isotropic model and an active contraction of the myocardium model. Pulsatile unloading is provided by the left ventricular assist device (LVAD), which is implanted between the aortic root and aortic arch. Support models (constant speed and co-pulse) were utilized to study the effect of pulsatile unloading on intraventricular flow and ventricular stress. The result indicates that the formation time of the vortex increases under pulsatile unloading. The area rate of high time-averaged wall shear stress (TAWSS) increased after pulsatile unloading. The area of the high oscillatory shear index (OSI) region (OSI > 0.375) was calculated for heart failure, constant speed, and co-pulse (9.9 cm(2), 9.6 cm(2), and 9.2 cm(2), respectively). The maximum value of the stress that reflects the level of stretch declined after pulsatile unloading (66.4 kPa, 30.9 kPa, and 21.3 kPa, respectively). Besides, pulsatile unloading impacts the maximum value of thickness at the ventricular wall (-0.75 mm, -1 mm, and -1.25 mm, respectively). The change ratios of the thickness are 10%, 14%, and 17%, respectively. In conclusion, pulsatile unloading contributes to the distribution of intraventricular flow and the formation time of the vortex. Co-pulse support significantly reduces the maximum value of the ventricular wall stress and the area of high stress on the ventricular wall. (C) 2019 Elsevier Ltd. All rights reserved.

Methylation signature genes identification of cancers occurrence and pattern recognition SCIE

In order to identify the signature genes of tumorigenesis, the pattern-recognition method was used to analyze the gene methylation (ME) data which included only normal and cancer samples and was collected from the TCGA (The Cancer Genome Atlas) database. Here, we analyzed the DNA methylation profiles of the six types of cancer and the ME signature genes for each cancer were selected by means of a combination of correlation, students t-test and Elastic Net. Modeling by support vector machine, the accuracy of ME signature genes can be as high as 98 % for training set and as high as 97 % for the independent test set, the recognition accuracy of stage I is more than 97 % for training set and more than 98 % for test set. Then, the common signature genes and common pathways emerging in multiple cancers were obtained. A functional analysis of these signature genes indicates that the identified signatures have direct relationship with tumorigenesis and is very important for understanding the pathogenesis of cancer and the early therapy.
