G-quadruplex (G4) DNA is present in human telomere oligonucleotide sequences. Oxidative damage to telomeric DNA accelerates telomere shortening, which is strongly associated with aging and cancer. Most of the current analyses on oxidative DNA damage are based on ds-DNA. Here, we developed a electrochemiluminescence (ECL) probe for enhanced recognition of oxidative damage in G4-DNA based on DNA-mediated charge transfer (CT), which could specifically recognize damaged sites depending on the position of 8-oxoguanine (8-oxoG). First, a uniform G4-DNA monolayer interface was fabricated; the G4-DNA mediated CT properties were examined using an iridium(III) complex [Ir(ppy)2(pip)]PF6 2 (pip)]PF 6 stacked with G4-DNA as an indicator. The results showed that G4- DNA with 8-oxoG attenuated DNA CT. The topological effects of oxidative damage at different sites of G4- DNA and their effects on DNA CT were revealed. The sensing platform was also used for the sensitive and quantitative detection of 8-oxoG in G4-DNA, with a detection limit of 28.9 fmol. Overall, these findings present a sensitive platform to study G4-DNA structural and stability changes caused by oxidative damage as well as the specific and quantitative detection of oxidation sites. The different damage sites in the G-quadruplex could provide detailed clues for understanding the function of G4-associated telomere functional enzymes.

一种基于铂铱介孔合金的NO电化学传感器及其制备方法属于电化学传感器技术领域。本文通过电化学沉积技术在电极表面原位生成介孔铂铱合金纳米颗粒(Pt‑Ir NPs),可实现在生理条件下准确快速地检测NO。介孔通过在电解液中添加辅助性表面活性剂来构建。此外,该传感器还具有线性范围宽、灵敏度高、检出限低、响应时间短、抗干扰能力强等优点,有望在生物医学分析检测领域发挥重要作用,并且具有商业化的潜力。

Here, we present a dual amplification strategy of electrochemiluminescence (ECL) based on aggregation-induced and photoelectron transfer (PET) inhibition using a designed ruthenium-based complex for the highly sensitive detection of nitric oxide (NO). A novel ruthenium(II) complex ([Ru(phen)2(DA-phen)]2+ , DA-phen: 5,6-diamino- 1,10-phenanthroline) was synthesized that uses the O-diaminophenyl group for specific recognition of NO and the ruthenium metal center with 1,10-phenanthroline as the luminophore. In the first step, the 1,10-phenanthro- line groups function as the aggregation part. Increasing the volume fraction of a poor solution (H2O: fw , v%) in the DMSO mixture system from 0% to 80% yielded an increase in the fluorescence (FL) and electro- chemiluminescence (ECL) intensities by 3- and 42-fold when compared with the FL and ECL intensities in pure DMSO, respectively. The second step involves a PET process. Reacting the complex with NO in physiological pH aqueous media under aerobic conditions to afford its triazole derivative, [Ru(phen)2(TA-phen)]2+ (TA-phen: 5,6- triazole-1,10-phenanthroline), inhibited the electron transfer process, yielding a remarkable further increase in the FL and ECL efficiencies of the Ru(II) complex. The ECL intensities was 86-fold higher in the presence of NO. This dual amplification strategy enabled the sensitive detection of NO from 1.8 x 10-10 to 1.8 x 10-5 M with a detection limit of 60 pM (S/N=3) and offered excellent specificity with no detectable interference by RNS and metal ions. Furthermore, selective NO detection in cells was achieved using this complex. Thus, this assay should be suitable for applications in environmental toxicology research and early clinical diagnosis.

In this paper, we systematically investigated the aggregation-induced electrochemiluminescence (AIECL) of the ruthenium(II) complex ([Ru(phen)2(phen-O2)](PF6)2, phen-O2: 5,6-diketone-1,10-phenanthroline), which was dissolved in solvents with different water/acetonitrile volume ratios. The fluorescence intensity of the system increased 11.5-fold when the proportion of water (a poor solvent) in the solvent mixture was increased to 90%, and the electrochemiluminescence intensity increased 16-fold when the proportion of water in the solvent was increased to 80%. Transmission electron microscopy and dynamic light scattering confirmed the aggregation of [Ru(phen)2(phen-O2)](PF6)2 molecules into nanoclusters. Aggregation-induced electrochemiluminescence properties were observed to increase with an increase in the proportion of water in H2O/CH3CN. Due to the excellent aggregation-induced luminescence performance of [Ru(phen)2(phen-O2)](PF6)2, the quantitative detection of dopamine was achieved with a good linear range of 100 pM-1uM and a limitation of detection of 33.3 pM. This simple and sensitive method based on the AIECL effect can be widely applied to the detection and analysis of other small biological molecules.

半胱胺作为电致化学发光共反应剂的应用,属于电致化学发光分析方法技术领域。将半胱胺作为电致化学发光共反应剂,三联吡啶钌(Ru(bpy)3Cl2)作为发光体,用于三联吡啶钌或半胱胺的电化学发光检测。与现有常用共反应剂(三正丙胺,TPrA)相比,该共反应剂具有较强的ECL增强、低生物毒性,未来可用于细胞检测等优势。实验结果表明:本发明的半胱胺作为三联吡啶钌电致化学发光共反应剂的应用,能够用于高效检测三联吡啶钌或半胱胺的含量,更重要的是,在一定浓度范围内,通过实验给出了半胱胺和三联吡啶钌在混合体系中的最佳终浓度。

As a key factor in cellular metabolic processes, nitric oxide (NO) is a challenging target for in situ real-time monitoring due to its transient property and short diffusion distance. Scanning electrochemical microscopy (SECM) has unique advantages in single-cell analysis, which can obtain the electrochemical current by scanning the cell surface with a tip microelectrode. In particular, it can further improved the electrochemical response by enhancing the interface properties of its tip. Here, an interface design strategy based on platinum single nano-particle (Pt NP) was developed, and fluorinated self-assembled monolayers (SAMs) were used to further improve its performance. This modified tip was used as an SECM probe for NO concentration monitoring and morphological imaging of single MCF-7 cells. It has the high sensitivity (164.7 mu A/mu M center dot cm(2)) and good selectivity for NO detection, which benefits from the efficient catalytic properties of Pt NPs and high mass transport and hydrophobic antifouling properties of the interface. Notably, it shows a superior performance in detecting the fluctuation of NO released by a single MCF-7-cell under the stimulation of cadmium (Cd), which demonstrates a promising method for using a single-particle-based tip in SECM applications.

基于S-Cd和N-Cd的亲和作用,利用一锅法制备了MoS_2-掺氮石墨烯(NG)新型纳米复合材料。采用方波溶出伏安法实现了Cd~(2+)的灵敏测定。在最优实验条件下,Cd~(2+)的溶出峰电流与Cd~(2+)的浓度在0.01~20μmol/L范围内呈线性关系(R~2=0.996),检出限为2.50 nmol/L。该传感器可用于监测实际样品中Cd~(2+)。

本文基于氧化石墨烯(GO)、电化学还原氧化石墨烯(ERGO)和氮掺杂石墨烯(NG)三种石墨烯材料修饰的电极制备了镉离子(Cd2+)电化学传感器.利用循环伏安法和差分脉冲伏安法分析检测Cd2+,系统的比较了不同石墨烯材料修饰电极的电化学性质及检测效果.结果表明GO修饰的传感器在灵敏度、检测限和可重复性方面优于ERGO和NG,说明了石墨烯上含氧基团的存在提高了Cd2+检测的灵敏度.
