Association analysis of BclI with benign lymphoepithelial lesions of the lacrimal gland and glucocorticoids resistance SCIE

AIM: To evaluate the relationship between gene polymorphism (BclI, ER22/23EK, N363S) and the occurrence, progression and sensitivity to glucocorticoid of lacrimal gland benign lymphoepithelial lesion (LGBLEL). METHODS: Clinical peripheral blood samples of LGBLEL patients and 10 normal volunteers were collected for DNA extraction and polymerase chain reaction sequencing to analyze single nucleotide polymorphism (SNP) genotypes. The lacrimal tissues of LGBLEL were surgically removed and made into paraffin sections for subsequent hematoxylin-eo sin (HE) and Masson staining analysis. The duration of disease and hormone use of LGBLEL patients from diagnosis to surgery were also analyzed. Meta-analysis follows PRISMA guidelines to conducted a systematic review of human studies investigating relationship between the NR3C1 BclI polymorphism and glucocorticoids (GCs) sensitivity. RESULTS: There was no association between ER22/23EK or N363S and the occurrence of LGBLEL or GCs sensitivity (P>0.05); BclI GC genotype was closely related to GCs resistance (P=0.03) as is the minor allele C (P=0.0017). The HE staining and Masson staining showed that the GC genotype of BclI remarkably slowed down the disease progression and reduced fibrosis (P<0.05), especially for GCs-dependent patients (P<0.0001). Meta-analysis showed that BclI was not significantly associated with GCs responsiveness. CONCLUSION: The LGBLEL patients who carry the NR3C1 BclI allele C may be more sensitive to GCs and associated with lower fibrosis and slower disease progression. The results may guide the clinical treatment strategy for the LGBLEL patients.

Development and Prospective Applications of 3D Membranes as a Sensor for Monitoring and Inducing Tissue Regeneration SCIE

For decades, tissue regeneration has been a challenging issue in scientific modeling and human practices. Although many conventional therapies are already used to treat burns, muscle injuries, bone defects, and hair follicle injuries, there remains an urgent need for better healing effects in skin, bone, and other unique tissues. Recent advances in three-dimensional (3D) printing and real-time monitoring technologies have enabled the creation of tissue-like membranes and the provision of an appropriate microenvironment. Using tissue engineering methods incorporating 3D printing technologies and biomaterials for the extracellular matrix (ECM) containing scaffolds can be used to construct a precisely distributed artificial membrane. Moreover, advances in smart sensors have facilitated the development of tissue regeneration. Various smart sensors may monitor the recovery of the wound process in different aspects, and some may spontaneously give feedback to the wound sites by releasing biological factors. The combination of the detection of smart sensors and individualized membrane design in the healing process shows enormous potential for wound dressings. Here, we provide an overview of the advantages of 3D printing and conventional therapies in tissue engineering. We also shed light on different types of 3D printing technology, biomaterials, and sensors to describe effective methods for use in skin and other tissue regeneration, highlighting their strengths and limitations. Finally, we highlight the value of 3D bioengineered membranes in various fields, including the modeling of disease, organ-on-a-chip, and drug development.

Long-term and daily use of molecular hydrogen induces reprogramming of liver metabolism in rats by modulating NADP/NADPH redox pathways SCIE

Molecular hydrogen (H-2) has emerged as a new therapeutic option in several diseases and is widely adopted by healthy people. However, molecular data to support therapeutic functions attributed to the biological activities of H-2 remain elusive. Here, using transcriptomic and metabolomic approaches coupled with biochemistry and micro-CT technics, we evaluated the effect of long-term (6 months) and daily use of H-2 on liver function. Rats exposed 2 h daily to H-2 either by drinking HRW (H-2 dissolved in H2O) or by breathing 4% H-2 gas showed reduced lipogenesis and enhanced lipolysis in the liver, which was associated with apparent loss of visceral fat and brown adipose tissue together with a reduced level of serum lipids. Both transcripts and metabolites enriched in H-2-treated rats revealed alteration of amino acid metabolism pathways and activation of purine nucleotides and carbohydrate biosynthesis pathways. Analysis of the interaction network of genes and metabolites and correlation tests revealed that NADP is the central regulator of H-2 induced metabolic alterations in the liver, which was further confirmed by an increase in the level of components of metabolic pathways that require NADP as substrate. Evidence of immune response regulation activity was also observed in response to exposure to H-2. This work is the first to provide metabolomic and transcriptomic data to uncover molecular targets for the effect of prolonged molecular hydrogen treatment on liver metabolism.

本发明公开了一种用于恢复TBI损伤后脑功能的氢分子治疗剂,所述治疗剂由氢气和氧气组成所述氢气体积百分比浓度为5%‑95%,氧气体积百分比浓度为5%‑95%,该治疗剂通过吸入给药可以减轻TBI后脑组织及外周系统神经炎症反应发挥神经保护作用,表现为外周血中白细胞、淋巴细胞、单核细胞、中性粒细胞以及小胶质细胞均有不同程度的降低,同时CD16阳性细胞减少,CD206阳性细胞进一步增多,并通过促进小胶质细胞M1‑to‑M2表型转化发挥其神经保护和治疗作用。

Effect of hydrogen intervention on refractory wounds after radiotherapy: A case report SCIE

BACKGROUND Patients with keloids who receive radiotherapy (RT) after surgery can develop refractory wounds that cannot be healed by the patient's own repair system. Such chronic wounds are uneven and complex due to persistent abscess and ulceration. Without external intervention, they can easily result in local tissue necrosis or, in severe cases, large area tissue resection, amputation, and even death. CASE SUMMARY This article describes the use of hydrogen to treat a 42-year-old female patient with a chronic wound on her left shoulder. The patient had a skin graft that involved implanting a dilator under the skin of her left shoulder, and then transferring excess skin from her shoulder onto scar tissue on her chest. The skin grafting was followed by two rounds of RT, after which the shoulder wound had difficulty healing. For six months, the patient was treated with 2 h of hydrogen inhalation (HI) therapy per day, in addition to application of sterile gauze on the wound and periodic debridement. We also performed one deep, large, sharp debridement to enlarge the wound area. The wound healed completely within 6 mo of beginning the HI treatment. CONCLUSION After HI therapy, the patient showed superior progress in reepithelializa Lion and wound repair, with eventual wound closure in 6 mo, in comparison with the previous failures of hyperbaric oxygen and recombinant bovine basic fibroblast growth factor therapies. Our work showed that HI therapy could be a new strategy for wound healing that is deaner, more convenient, and less expensive than other therapies, as well as easily accessible for further application in clinical wound care.

氢气以其安全、有效、渗透性强、代谢产物只有水等特点,逐渐进入医学研究者和临床工作者的视野,并在近年来快速发展.大量研究证明,氢气对于包括肿瘤、炎症损伤、代谢疾病、神经性疾病等百余种疾病具有潜在的治疗作用.2020年新型冠状病毒肺炎(新冠肺炎)疫情全球大流行期间,氢气在新冠肺炎辅助治疗中也崭露头角.对氢气在生物医学的动物学、细胞学和临床研究领域的研究进展进行了总结,并主要综述了氢气在代谢性疾病、神经退行性疾病、急救和创伤医学以及肿瘤领域的研究进展,以期为氢气在生物医学领域的应用研究提供参考.

布鲁氏菌病是世界公认的危害较为严重的人畜共患传染病之一.由于该病防治相对困难,在我国发病形势尤为严峻.尽管我国对于布鲁氏菌病防治采取了积极的措施,但仍然面临着巨大挑战.因此,认清布鲁氏菌病的病原学特征、传播途径以及自然史等流行病学特征,并了解当前布鲁氏菌病的诊断、治疗以及预防现状,对于预防布鲁氏菌病的发生、减少布鲁氏菌病对农业及人类造成的健康财产损害有着重要意义.

本发明公开了氢分子在制备用于调节肠道菌群的药品或食品中的用途,氢分子的载体形式为富氢液体,富氢液体中氢气浓度不低于1.0mmol/L。本发明还提供了一种用于调节肠道菌群的药品或食品,为氢气浓度不低于1.0mmol/L的富氢液体。该富氢液体能调节肠道菌群稳态,尤其是可显著增加健康大鼠肠道菌群中益生菌乳酸杆菌属(Lactobacillus)和双歧杆菌属(Bifidobacterium)的相对丰度,优化肠道菌群结构,还可提高粪便即肠道内容物中乙酸和丙酸等短链脂肪酸的含量,还可调节肠道菌群的产氢能力,影响氢气产量。
